Изучить влияние ГКСК на рост, жизнеспособность, пролиферативную активность и пул стволовых клеток рака шейки матки in vitro.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Клетки линии HeLa инкубировали с коммерчески доступной ГКСК (торговое название Сферо®ГЕЛЬ в двух вариантах исполнения – Medium и Light) в течение 24–72 ч. Определяли общее количество опухолевых клеток, их жизнеспособность методом МТТ, суммарную долю клеток в фазах S + G2 + M методом проточной цитометрии, а также относительное и абсолютное количество опухолевых стволовых клеток (ОСК), идентифицированных по способности к выведению флуоресцентного красителя Хёхст 33342 из клеток (метод SP) и экспрессии CD133, после инкубации с ГКСК в двух разведениях и в контрольных образцах.</p></sec><sec><title>Результаты</title><p>Результаты. При использовании ГКСК-Мedium в разведении 1/5 (наименьшем из изученных разведений) зарегистрировано уменьшение общего числа опухолевых клеток на 13 % от контрольного уровня в течение всего периода наблюдения (p &lt; 0,05). Этот эффект сопровождался параллельным снижением жизнеспособности клеток и уменьшением пролиферативной активности в первое время после начала воздействия по сравнению с контролем (p &lt; 0,05). Отмечена тенденция к уменьшению абсолютного количества ОСК, выявляемых независимо методами SP и иммунофенотипирования по CD133 после 72-часовой инкубации с ГКСК-Мedium 1/5, в 1,3 раза по сравнению с контролем. Сходные, но кратковременные или менее выраженные эффекты показаны для ГКСК-Medium в большем разведении 1/20 и ГКСК-Light во всех разведениях в отношении общего количества опухолевых клеток и размера пула ОСК.</p></sec><sec><title>Заключение</title><p>Заключение. Полученные результаты доказывают отсутствие стимулирующего действия ГКСК-Мedium и ГКСК-Light как на общую массу опухолевых клеток линии HeLa, так и на субпопуляцию ОСК в условиях in vitro, и показывают перспективность дальнейшего проведения доклинических и клинических исследований по применению ГКСК в реабилитационных программах онкологических больных для лечения атрофических и/или лучевых вагинитов различной степени тяжести.</p></sec></abstract><trans-abstract xml:lang="en"><p>   Understanding the effects on tumor cells underlies possible practical use of collagen- containing composition (HCCC) for the treatment of radiation therapy related complications.</p><sec><title>   Purpose of the study</title><p>   Purpose of the study. Is to investigate the effects of heterogeneous HCCC on growth, viability, proliferative activity and stem cell pool of cervical cancer in vitro.</p></sec><sec><title>   Materials and methods</title><p>   Materials and methods. HeLa cells were incubated with commercially available HССC (trademark Sphero®GEL in two versions: Medium and Light) for 24–72 h. We determined the total number of tumor cells, their viability by MTT assay, the total proportion of cells in S + G2 + M phases by flow cytometry, as well as the relative and absolute number of cancer stem cells (CSCs) identified by the ability to remove the fluorescent dye Hoechst 33342 from cells (SP method) and CD133 expression, after incubation with HCCC in two dilutions and in control samples.</p></sec><sec><title>   Results</title><p>   Results. When using HCCC–Medium in a dilution of 1/5 (the lowest of the studied dilutions), decrease in the total number of tumor cells by 13 % from the control level was recorded during the entire observation period (p &lt; 0.05). This effect was accompanied by parallel decrease in cell viability and decrease in proliferative activity in the first time after the onset of exposure compared to control (p &lt; 0.05). The tendency towards decreases in the absolute number of CSCs, which were independently detected by SP and CD133 immunophenotyping methods after 72-hour incubation with HCCC–Medium 1/5, was observed. The number of SP and CD133+ cells decreased by 1.3 times compared to control.
