Роль митохондриального стероидогенеза в прогрессии аденокарциномы эндометрия (АЭ), особенно на фоне сахарного диабета 2-го типа (СД2), остается малоизученной.</p><sec><title>Цель исследования</title><p>Цель исследования. Определение концентраций стероидных гормонов и их рецепторов в митохондриальной фракции ткани АЭ степеней дифференцировки G1–G3 в сравнении с интактным эндометрием, а также оценка модифицирующего влияния СД2 на данные показатели.</p></sec><sec><title>Пациенты и методы</title><p>Пациенты и методы. В митохондриях, выделенных из опухолевой ткани 92 больных АЭ (G1–G3) и морфологически нормального эндометрия (контроль) методом дифференциального центрифугирования, стандартными ИФА наборами определяли содержание гормонов: эстрадиола (Е2), эстриола (Е3), эстрона (Е1) и их рецепторов (REα, REβ), тестостерона (Т) и его рецептора (RA), прогестерона (Р4) и его рецептора (RP4), кортизола и холестерина. Статистическая обработка выполнена с использованием программы Statistica 10.</p></sec><sec><title>Результаты</title><p>Результаты. В митохондриях АЭ G1–G2 уровень эстрадиола в 2 раза выше, чем в контроле, тогда как при G3 была повышена концентрация эстриола в среднем в 2 раза. В митохондриях всех опухолей был повышен уровень Т и кортизола в 1,6 раза и более чем в 2 раза соответственно. Содержание Р4 было повышено в митохондриях АЭ G1–G2 в 2,3–3,8 раза, но снижено в 4,3–5,9 раза, по сравнению с митохондриями контроля. В митохондриях всех опухолей были повышены RA и рецепторы эстрогенов, со снижением коэффициента REα/REβ в 1,7–3,6 раза, уровень RP4 повышен в митохондриях G1–G2 только при СД2, и при G3 независимо от СД2. При СД2 выявлено накопление холестерина в митохондриях всех опухолей.</p></sec><sec><title>Заключение</title><p>Заключение. В митохондриях АЭ независимо от степени дифференцировки опухоли и СД2 повышены Т, кортизол и рецепторы половых гормонов, что отражает метаболическую перестройку органелл. При G1–G2 доминирует эстрадиол, при G3 – эстриол, что может влиять на агрессивность опухоли. СД2 ассоциирован с накоплением холестерина в митохондриях, что может объяснять более тяжелое течение АЭ у таких пациенток.</p></sec></abstract><trans-abstract xml:lang="en"><p>Mitochondria play a key role in cellular metabolism, steroid hormone synthesis, and the regulation of redox homeostasis. However, the role of mitochondrial steroidogenesis in the progression of endometrial adenocarcinoma (EA), particularly in the presence of type 2 diabetes mellitus (T2DM), remains poorly understood.</p><sec><title>Purpose of the study</title><p>Purpose of the study. To determine the concentrations of steroid hormones and their receptors in the mitochondrial fraction of G1–G3 endometrial adenocarcinoma tissue compared with intact endometrium and to evaluate the modifying effect of type 2 diabetes mellitus on these parameters.</p></sec><sec><title>Patients and methods</title><p>Patients and methods. Mitochondria were isolated by differential centrifugation from tumor tissue obtained from 92 patients with endometrial adenocarcinoma (G1–G3) and from morphologically normal endometrium (control group). Standard enzyme-linked immunosorbent assay (ELISA) kits were used to determine the concentrations of estradiol (E2), estriol (E3), estrone (E1) and their receptors (ERα, ERβ), testosterone (T) and the androgen receptor (AR), progesterone (P4) and the progesterone receptor (PR), as well as cortisol and cholesterol. Statistical analysis was performed using Statistica 10 software.</p></sec><sec><title>Results</title><p>Results. In the mitochondria of G1–G2 endometrial adenocarcinomas, the estradiol level was approximately twofold higher than in the control group, whereas G3 tumors demonstrated an approximately twofold increase in estriol concentration. Testosterone and cortisol levels were elevated in the mitochondria of all tumors by 1.6-fold and more than twofold, respectively. Progesterone content was increased by 2.3–3.8-fold in the mitochondria of G1–G2 tumors but decreased by 4.3–5.9-fold in comparison with the control mitochondria. The levels of androgen and estrogen receptors were elevated in the mitochondria of all tumors, whereas the ERα/ERβ ratio decreased by 1.7–3.6-fold. Progesterone receptor levels were increased in G1–G2 mitochondria only in patients with T2DM, whereas in G3 tumors they were elevated regardless of T2DM status. Type 2 diabetes mellitus was associated with cholesterol accumulation in the mitochondria of all tumors.</p></sec><sec><title>Conclusion</title><p>Conclusion. Regardless of tumor differentiation grade and the presence of type 2 diabetes mellitus, endometrial adenocarcinoma mitochondria are characterized by increased levels of testosterone, cortisol, and sex hormone receptors, reflecting profound metabolic reprogramming of these organelles. Estradiol predominates in G1–G2 tumors, whereas estriol is the dominant estrogen in G3 tumors, which may contribute to increased tumor aggressiveness.
