Создание Patient Derived Xenograft (PDX)‑модели немелкоклеточного рака легкого на иммунодефицитных мышах, адаптированной к росту на иммунодефицитных мышах.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Для проведения данной работы использовали опухолевый материал от 14 доноров, имплантированный подкожно 132 иммунодефицитным мышам линии Balb/c Nude. Ксенографты поддерживали до 3‑го пассажа. Для проверки чувствительности созданных моделей к цисплатину использовали PDX от 3‑х пациентов 3‑го пассажа. Был проведен гистологический анализ и генетическое исследование на наличие мутаций в гене EGFR донорских опухолей от 3‑х пациентов и соответствующих ксенографтов 3‑го пассажа.</p></sec><sec><title>Результаты</title><p>Результаты. В нашей работе заметный рост PDX отмечался уже на 8‑е сутки после перевивки опухолевого материала. При этом успешное приживление ксенографтов отмечалось у 21 мыши из 42, что составляет 50 %. Данные показатели являются довольно успешным результатом. Проведенный сравнительный гистологический анализ опухолевого материала от 3‑х пациентов показал сохранение PDX‑моделями первоначального гистотипа. Также нами была продемонстрирована идентичность полученных ксенографтов от 3‑х пациентов донорской опухоли в аспекте анализа наличия мутаций в гене EGFR, что доказывает ценность данных PDX‑моделей для использования для доклинических исследований веществ с потенциальной противоопухолевой активностью. В исследовании чувствительности полученных ксенографтов к цисплатину‑цитостатику, в ксенографтах, полученных от двух пациентов из 3‑х, наблюдалось статистически значимое замедление скорости роста по сравнению с контролем.</p></sec><sec><title>Заключение</title><p>Заключение. Созданные нами PDX‑модели могут быть рекомендованы в качестве тест‑систем для доклинических исследований эффективности новых фармакологических субстанций с потенциальной противоопухолевой активностью.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Purpose of the study</title><p>Purpose of the study. Was was the creation of a Patient Derived Xenograft (PDX) model of non‑small cell lung cancer in immunodeficient mice adapted to growth in immunodeficient mice.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The study was performed using the tumor material from 14 donors implanted subcutaneously to 132 immunodeficient Balb/c Nude mice. Xenografts were maintained until the third passage. PDXs in the third passage from 3 patients were used to assess the model sensitivity to cisplatin. A histological analysis and genetic tests for the presence of EGFR mutations were performed for donor tumors from 3 patients and the corresponding xenografts in the third passage.</p></sec><sec><title>Results</title><p>Results. We observed a noticeable PDX growth already on the 8th day after the tumor material implantation. Successful xenograft engraftment was noted in 21 of 42 mice (50 %), which were rather successful results. A comparative histological analysis of tumor material from 3 patients showed that the PDX models retained the original histotype. We also demonstrated the identity of the EGFR mutations in the established xenografts from 3 patients and the donor tumors, which proved the value of these PDX models for preclinical studies of substances with potential antitumor activity. The analysis of the xenograft sensitivity to cytostatic cisplatin showed a statistically significant decrease in the growth rate in the xenografts obtained from 2 out of 3 patients, in comparison with the control.</p></sec><sec><title>Conclusions</title><p>Conclusions.
